Pick the anchor first
Museo de Antioquia is the obvious anchor for travelers interested in Botero and central Medellín. MAMM is a different experience: contemporary art, a different neighborhood and a more modern city feel. Casa de la Memoria is history and memory rather than an art-first stop.
Trying to make all three mandatory in four hours usually creates transportation instead of culture.
A Botero-focused half day
Pair Plaza Botero with Museo de Antioquia, then use the remaining time for a short Centro context walk or a nearby coffee stop. This is compact and makes sense geographically.
If the museum is the main reason for booking, give it enough time. Do not advertise a museum and then rush guests through it to protect a checklist.
A contemporary-art half day
MAMM can anchor a more modern route. Pair it with a neighborhood/architecture stop or a relaxed coffee break rather than crossing the city for a second major museum.
This route works well for repeat visitors who have already seen the classic central sights.
History and memory
Casa de la Memoria can be paired with broader city-history context. For guests interested in the Escobar era, conflict or transformation, it provides a more grounded frame than turning the entire day into narco sightseeing.

The closure problem
Museums and attractions have closure days, holidays and changing hours. A private planner should verify the official schedule for the actual date instead of assuming a generic weekly pattern.
